Please note that the 20x Stock is only provided in the summer. This is because the high concentration causes crystals to precipitate at low temperatures, and this product requires filter sterilisation at a relatively higher room temperature for several hours, which condition is difficult to achieve at lower temperatures.
For cell lysate generation and protein extraction
S30A/B provides additional glutamate compared to S30, which can support the tricarboxylic acid cycle to produce more energy molecules (e.g. ATP) and provide a longer active window for cell lysates.
Per 1000mL of S30B buffer
Tris Base 0.606g
Mg-Glutamate 5.44g
K-Glutamate 12.194g
pH 8.2
Preparation
Dilute 1 volume of S30B buffer 10x stock with 9 volumes of Million-Q water and use directly.
Or dilute 1 volume of S30B buffer 20x stock with 19 volumes of Million-Q water and use directly.
Before use, add 1 ml of 1 M DTT (2mM, final concentration) to 500 ml S30B buffer.
Please contact us if a customised HEPES version of S30B Buffer is needed.
*Heat the solution in a water bath to over 30°C if precipitates appear at the bottom of the solution. DO NOT use until the precipitate is completely dissolved.
*Because the solution is prone to form minor precipitates of Mg(OH)2 / MgCO3 at high pH, the original 50x stock product has been discontinued and updated to 10x stock.
