PRAS Dilution Buffer is a pre-reduced, anaerobically packaged mineral-salt buffer for serial dilution of obligate anaerobes before plating or colony enumeration.
Overview
PRAS Dilution Buffer - PRAS Dilution Buffer is a pre-reduced, anaerobically packaged mineral-salt buffer for serial dilution of obligate anaerobes before plating or colony enumeration.
The product is a companion to GMExpression's YCFA and anaerobic culture-media line: the same customer who needs anaerobic media often needs a reliable pre-reduced dilution system that does not introduce oxygen during sample handling.

Use and Applications
- Serial dilution of anaerobic bacteria from 10-1 to 10-8 before plating
- Anaerobic microbiology and FMT sample processing
- Holding and dilution of oxygen-sensitive cultures before enumeration
- Distributor bundles with YCFA, chopped-meat media, and anaerobic preparation accessories

Composition
| Component | Amount / concentration | Function |
|---|---|---|
| KH2PO4 / K2HPO4 | Near-neutral phosphate buffer | pH buffering |
| NaCl, ammonium sulfate, MgSO4, CaCl2 | Low-millimolar mineral salts | Ionic strength and cofactor background |
| L-cysteine-HCl and sodium thioglycolate | Reducing-agent pair | Maintains low redox potential |
| Resazurin | 0.001% | Oxygen ingress indicator |
| Pre-boiled, N2-sparged water | q.s. to final volume | Low-oxygen solvent |
Instruction
1Verify the tube or bottle is colourless before use. Pink colour indicates oxygen ingress and the unit should be discarded.
2Withdraw liquid through the septum using a sterile syringe; do not open the vessel to air.
3Prepare serial dilutions using anaerobic technique, mixing gently after each transfer.
4Use any pierced unit within 24 hours unless validated in-house for longer use.
Note
- The redox-indicator behaviour is intentionally visible. It gives the customer a simple release-at-use check that ordinary aerobic dilution blanks cannot provide.
Cautions
Caution. Do not use pink or partially pink units for strict anaerobe work.
Caution. Needle and septum handling should follow institutional sharps and anaerobic-culture safety procedures.
Storage and Expiry · Safety
- Store at 15-25 deg C away from direct sunlight unless otherwise specified on the label.
- Initial released shelf life: 12 months pending real-time stability validation; target shelf life 24 months.
- Transport. Ships at ambient temperature; no cold chain required. Protect from freezing and from temperatures above 40 deg C.
- Handling and PPE. Wear a laboratory coat, nitrile gloves, and safety glasses for routine handling. Low chemical hazard; follow institutional anaerobic-culture and (for PRAS) sharps/septum handling procedures. Consult the Safety Data Sheet before use.

Frequently Asked Questions
Q1. Why not make ordinary PBS anaerobic at the bench?
Brief gas flushing does not reliably remove oxygen or maintain a reduced redox state during serial dilution. PRAS blanks are reduced, sealed, and indicator-controlled.
Q2. What does pink colour mean?
Pink resazurin indicates oxygen exposure. For obligate anaerobe dilution, discard the affected unit.
