Overview

Antifade Mounting Medium, Mowiol-Based, +/- DAPI - This Mowiol-based Antifade Mounting Medium is designed for fluorescence microscopy workflows requiring refractive-index matching, coverslip mounting, and protection against photobleaching.

The formulation is based on public-domain Mowiol-DABCO mountant chemistry and is offered with or without DAPI for nuclear counterstaining.

Instruction

1Bring mountant to room temperature and mix gently before use.
2Apply a small drop to the specimen and lower coverslip slowly to minimise bubbles.
3Allow curing according to the imaging protocol before sealing or long-term storage.
4Protect mounted slides from light.

Note

  • The DAPI-containing variant is convenient for standard fixed-cell imaging; choose the no-DAPI variant where nuclear staining is not desired or another channel is used.

Cautions

Caution. DAPI is a DNA-intercalating dye and a suspected mutagen. Avoid skin contact and ingestion, wear gloves, and dispose of DAPI-containing material as hazardous waste. The no-DAPI variant does not carry this hazard.
Caution. Not designed for live-cell imaging.

Storage and Expiry · Safety

  • Store at 2-8 deg C protected from light.
  • DAPI variant may be supplied frozen or refrigerated depending on final stability validation.
  • Transport. Ships at ambient temperature in insulated packaging (DAPI variant frozen/refrigerated and light-protected); place at the labelled storage temperature on receipt.
  • Handling and PPE. Wear a laboratory coat, nitrile gloves, and safety glasses. The mountant contains glycerol, Mowiol, Tris and DABCO (low hazard); the DAPI variant additionally requires mutagen-handling precautions and hazardous-waste disposal. Consult the Safety Data Sheet before use.

Q1. Is this a hard-set mountant?
It is a liquid-curing Mowiol-based mountant. Curing behaviour should be validated against the customer's coverslip and imaging workflow.
Q2. Can DAPI concentration be customised?
Yes. DAPI-free, 1 ug/mL DAPI, and custom counterstain variants are feasible.
