Trypan Blue 0.4% Solution - Trypan Blue 0.4% Solution is a routine cell-counting reagent used to distinguish viable cells from membrane-compromised cells by dye exclusion.

Instruction

1Mix cell suspension 1:1 with Trypan Blue solution unless the counter protocol specifies otherwise.
2Incubate briefly at room temperature.
3Count unstained viable cells and blue non-viable cells according to the instrument or hemocytometer method.
4Dispose of dye-containing waste according to institutional procedures.

Note

  • Trypan Blue can underestimate late-stage viability in some fragile cell types. Pair with protocol-specific viability controls where needed.

Cautions

Caution. Suspected carcinogen / reproductive hazard - Trypan Blue (CAS 72-57-1) is a benzidine-based azo dye that is a suspected human carcinogen and has shown teratogenic/reproductive effects in animal studies. Avoid all skin contact, ingestion, and inhalation of aerosols. Treat spills and stained consumables as hazardous chemical waste.
Caution. Not intended for in vivo use or therapeutic applications.

Storage and Expiry · Safety

  • Store at room temperature or 2-8 deg C protected from strong light.
  • Unopened shelf life target: 24 months after validation.
  • Transport. Ships at ambient temperature; no cold chain required. Protect from freezing and from temperatures above 40 deg C.
  • Handling and PPE. Wear a laboratory coat, nitrile gloves, and safety glasses; work in a well-ventilated area and avoid generating aerosols. Dispose of dye-containing waste via the hazardous-waste stream, not the sink. Consult the Safety Data Sheet before use.
