Overview

Ponceau S Staining Solution - Ponceau S Staining Solution is the standard rapid, reversible stain for confirming protein transfer to PVDF or nitrocellulose membranes before blocking and antibody incubation.

It is simple chemistry but high practical value: a failed transfer can be detected before the laboratory spends time and antibody on a blot that cannot succeed.

Use and Applications

  • Western blot transfer verification
  • Protein lane visualisation before blocking
  • Membrane cutting and loading-control checks
  • SDS-PAGE and Western blot bundle companion product

Instruction

1Cover membrane with enough stain to wet fully.
2Rock gently until protein bands are visible, usually 1-5 minutes.
3Image or mark lanes as needed.
4Destain with water, TBS, or TBST before blocking.

Note

  • Ponceau S staining is reversible and compatible with most downstream Western blot workflows after thorough destaining.

Cautions

Caution. Contains acetic acid. Avoid inhalation and eye contact.
Caution. Do not allow membranes to dry during staining or destaining unless the protocol explicitly permits it.

Storage and Expiry · Safety

  • Store at room temperature away from strong light.
  • Unopened shelf life target: 36 months after validation.
  • Transport. Ships at ambient temperature; no cold chain required. Protect from freezing and from temperatures above 40 deg C.
  • Handling and PPE. Wear a laboratory coat, nitrile gloves, and safety glasses for routine handling. Contains acetic acid; corrosive/irritant vapour. Handle in a well-ventilated area or fume hood; avoid eye contact and inhalation. Consult the Safety Data Sheet before use.

Frequently Asked Questions

Q1. Will Ponceau S interfere with antibodies?
Not when fully destained before blocking. Residual stain should be washed out.
Q2. Can the stain be reused?
Yes, if uncontaminated, but discard when signal weakens or particulates appear.
