Murashige and Skoog medium without Agar & Sucrose

250g, fine powder, up to make 50L liquid media.

Murashige and Skoog medium (MS, MSO/MS0, MS Basal media) is a plant growth medium used for the plant cell culture.

 

Applications

The principal component of In-vitro plant cultivation.

Plant tissue culture media like calli induction, shoot regeneration, and root formation.

For the germination of seeds of different plant species like Arabidopsis thaliana, and Nicotiana benthamiana, under sterile growth conditions.

Preparation

Suspend 4.74 g of the medium into distilled water. Mix well and make up to 1000ml. Heat with continuous stirring and aliquot into tubes or flasks. Sterilise in the autoclave at 121°C for 15 minutes.

Note: 

- The original pH is 3.51. Adjust pH as necessary to meet performance criteria.

- There are traces of insolubles at room temperature.

^ During preparation, avoid naked flames and do not mix the basal salt mixture with reducing substances vigorously.

 

Storage

Media powder is extremely hygroscopic and should be protected from atmospheric moisture. If possible, the unused powder should be stored in a sealed box with silica desiccant.

Note:  The colour of the inner sealing cap and the container body may turn yellow-brown during long-term storage. This is because the colour masterbatches in the HDPE have been oxidised, which will not affect the use of the medium.
