Preparation
Suspend 3.18g of the S&H medium WO Vitamins (or 4.19g S&H medium with Vitamins) into MQ or double-distilled water. Heat with continuous stirring till all dissolved. Make the volume to 1000ml. Adjust the medium to the desired pH using 1M HCl/NaOH and aliquot into tubes or flasks. Autoclave at 121°C for 15 minutes (the S&H media with Sucrose should be autoclaved at 115°C for 20 minutes or might turn dark at over 115°C).
During preparation, avoid naked flames, and DO NOT vigorously mix the basal salt mixture with reducing substances such as sucrose in powder form, which may cause an explosion.
Note
- DO NOT intensively hit the medium powder, as it contains ammonium and nitrate which may cause an explosion.
- This mixture contains a large amount of strong oxidising ingredients. We pre-packed the powder in a PP bag, which you can remove after opening. If the powder directly contacts the bottle, the surface of the plastic bottle and cap may turn yellow/brown after long-term storage, which will not affect the medium.
- If possible, the unused powder should be stored in a sealed glass reagent bottle and refrigerated at 2~8°C.
- When the liquid media is prepared, please store it at 2~8°C, the shelf life of such a stock can be several years as all ingredients and elements are in a stable inorganic salt form.
