    Use

    Columbia Agar Base is a nutrient-rich medium for the cultivation of fastidious organisms, commonly used as a base for Blood Chocolate Agar. It can also be used as a selective isolation medium by the addition of antimicrobial agents, suitable for the isolation and cultivation of a wide variety of microorganisms that are difficult to grow, like Neisseria meningitidis,Neisseria gonorrhoeae and Haemophilus influenzae. By adding blood, it can be used to determine haemolytic reactions, while the majority of common pathogenic bacteria grow well without the addition of blood. 

    Pancreatic digest provides general nitrogen, vitamins, minerals and amino acids, while yeast extract and maize starch are a source of vitamins essential for bacterial growth. Sodium chloride provides essential electrolytes for transport and osmotic balance. Blood is another source of growth factors for microorganisms and is the basis for determining haemolytic reactions.

    Preparation

    -       Dissolve 44 g of mixture A in 900~950 ml of MQ water in a closed container.

    -       Autoclave at 121°C for 15 minutes.


    [For making Blood Agar]

    -       Cool the Base: Cool the sterile molten agar base to 45–50°C, typically in a water bath to maintain a consistent temperature and prevent premature solidification.

    -       Add Blood: Aseptically add 5-10% (v/v, final) sterile defibrinated sheep/horse blood that has been pre-warmed to room temperature.

    -       Mix and Pour: Mix gently by swirling the flask or bottle, avoiding air bubbles. Quickly dispense the mixture into sterile Petri dishes.

    -       Solidify and Store: Allow the plates to solidify completely at room temperature (around 30 minutes to an hour) before inverting and storing them in sealed plastic bags at 2–8°C to prevent drying and condensation.


    [For making Chocolate Agar]

    -       Cool the Base: Cool the sterile molten agar base to 80°C, typically in a water bath to maintain a consistent temperature and prevent premature solidification.

    -       Add Blood: Aseptically add 10% (v/v, final) sterile defibrinated sheep/horse blood that has been pre-warmed to room temperature. Keep at 80°C for 15 minutes to lyse red blood cells, releasing hemoglobin.

    -       Mix and Pour: Mix gently by swirling the flask or bottle, avoiding air bubbles. Quickly dispense the mixture into sterile Petri dishes.

    -       Solidify and Store: Allow the plates to solidify completely at room temperature (around 30 minutes to an hour) before inverting and storing them in sealed plastic bags at 2–8°C to prevent drying and condensation.


    Note:

    It is strongly recommended to operate within a clean bench when preparing the solution & agar plate.


    When ordered with defibrinated blood, this item is shipped with 2.8 kg of dry ice for international deliveries and 1.3 kg of ice packs for domestic shipments.

    For international customers, we recommend sourcing defibrinated sheep or horse blood locally whenever possible to minimise the risk of customs clearance delays, which are common when importing animal blood products.
