Overview

SSC Buffer, 20x Stock, pH 7.0 - SSC Buffer is the standard high-salt citrate buffer base for nucleic-acid hybridisation and wash stringency control. A 20x stock lets users prepare 0.1x-6x working solutions depending on probe, target, and temperature conditions.

This candidate is a workflow-extension SKU for customers working in FISH, RNA-FISH, Southern blot, Northern blot, and hybridisation-chain-reaction protocols.

Instruction

1Dilute the 20x stock with ultrapure water to the protocol-specified working strength.
2Typical wash strengths range from 0.1x for high stringency to 6x for low stringency.
3Warm and mix if salt crystallisation is observed after cold storage.

Note

  • Hybridisation stringency is controlled by SSC concentration, formamide content, temperature, and probe length. Do not change SSC concentration without considering the whole protocol.

Cautions

Caution. High-salt solutions can crystallise during cold storage; fully redissolve before use.
Caution. Citrate chelates divalent cations and may interfere with metal-dependent downstream steps.

Storage and Expiry · Safety

  • Store at room temperature.
  • Unopened shelf life: 24 months from manufacture after QC validation.
  • Transport. Ships at ambient temperature; no cold chain required. Protect from freezing and from temperatures above 40 deg C.
  • Handling and PPE. Wear a laboratory coat, nitrile gloves, and safety glasses for routine handling. The formulated buffer presents no significant hazard under normal use; avoid ingestion and eye contact. Consult the Safety Data Sheet before use.
