Overview

Tyrode's Solution, Modified, 10x Stock - Tyrode's Solution is a classical physiological salt solution for cardiac muscle, smooth muscle, and platelet biology. The GMExpression candidate is a 10x modified carbonate-free stock designed for shelf stability and fast point-of-use preparation.

The product sits naturally in the Physiological Perfusate Family beside KHB and aCSF, giving pharma and academic physiology laboratories a consistent domestic supplier for three common perfusates.

Use and Applications

  • Cardiac safety pharmacology and isolated tissue bath experiments
  • Smooth muscle physiology studies
  • Platelet aggregation and coagulation-adjacent workflows
  • General physiological salt-solution preparation where Tyrode's composition is specified

Instruction

1Dilute 1 volume of 10x stock with 9 volumes of sterile ultrapure water (0.1μm filterated MQ water).
2If the bicarbonate/carbogen protocol is required, add NaHCO3 after dilution and equilibrate with carbogen.
3Warm to the protocol-specified temperature before use in organ bath or platelet assays.
4Use diluted 1x solution within 24 hours.

Note

  • Tyrode's formulations vary by discipline. This page describes the modified shelf-stable stock; custom Ca-free, HEPES-only, or bicarbonate-forward variants are feasible.

Cautions

Caution. Do not store diluted bicarbonate-containing Tyrode's for long periods.
Caution. Check calcium and magnesium requirements carefully for platelet and smooth-muscle protocols.

Sterile Filtration - Mycoplasma-Reduction Grade

Because this solution is used with living mammalian cells and tissue, it is sterilised through a 0.1 μm membrane rather than the conventional 0.22 μm sterilising grade. Mycoplasma are deformable enough to pass through 0.22 μm pores; where a 0.1 μm filteration gives a substantially lower mycoplasma margin, and each lot is released mycoplasma-test-negative by PCR.

Storage and Expiry · Safety

  • Store 10x stock at 2-8 deg C.
  • Unopened shelf life: 24 months from manufacture after QC validation.
  • Transport. Ships at ambient temperature in insulated packaging; brief transit excursions are tolerated. Refrigerate at 2-8 deg C on receipt and protect from freezing.
  • Handling and PPE. Wear a laboratory coat, nitrile gloves, and safety glasses for routine handling. The salt stock presents no significant chemical hazard; maintain aseptic technique. Consult the Safety Data Sheet before use.

Frequently Asked Questions

Q1. Is this the same as KHB?
No. Both are physiological salt solutions, but Tyrode's has a different ion profile and is used for different tissue systems.
Q2. Can HEPES-free Tyrode's be supplied?
Yes. A bicarbonate/carbogen-only variant is feasible as a custom formulation. Quotation separately via support@gmexpression.com
