Ultra-flux His-tag Affinity Microsphere (Ni-NTA Ultra-Flux Affinity Microsphere Gel)

Research use, for the purification of proteins with His-tag.

The default particle size is 50μm and the pore size is 800 Å. The particle size of 15/30/100/200/500μm and pore size 300/500/800Å are available upon request.

Ni-NTA Ultra-Flux Affinity Microsphere for the purification of proteins with His-tag. The microsphere core of this affinity gel is not traditional agarose but high-density polyacrylate (HDPA).

  • PA microspheres have a more uniform particle size (50μm±5%) distribution than agarose microspheres
  • Shelf life up to 5 years, no need for 4 degree Celsius environment, only need room temperature storage
  • High protein binding capacity, dynamic binding capacity up to ~90mg/ml ±10%
  • Target protein elution peaks are much sharper than traditional agarose microsphere affinity gels
  • Tolerates extreme affinity/elution flux and pressures (faster affinity/elution rates allow higher pressures to be applied to the gel without worrying about microsphere fragmentation)
  • The higher binding strength of the ligands cross-linked on the microsphere surface, leads to high reusability of the gel, which can be reused at least 20 times (up to 30 times in practice)
  • Less toxicity compared to conventional agarose microsphere due to the low probability of ligand elution.


Ultra-Flux Affinity Microsphere (UFAM) is an advanced protein purification filler with a high flow rate, high binding capacity and high resolution. This chromatography medium is prepared by using high-density polyacrylate microspheres as the substrate and various functional ligand groups or ligands cross-linked on its surface. UFAM can purify target proteins with His tag directly from cell lysates and is especially suitable for purifying target proteins directly from supernatants (e.g. cell culture supernatants) containing metal chelators, such as amino acids, EDTA and other components.


High Flow Rate

UFAM has high mechanical stability and uniform particle size, resulting in better chromatographic characteristics, excellent compression resistance and less chance of blockage. It can withstand pressures up to 10 MPa during operation and supports flow rates up to 1800 cm/hour (recommended flow rates 150-700 cm/hour).


High binding capacity

UFAM is a porous microsphere with a uniform distribution of pore and particle size, and its specific surface area is up to >500m2/g. Chromatographic filler prepared with it has a high density of functional groups and an extraordinary dynamic binding capacity, which is more than twice that of conventional agarose microsphere cores.


High resolution

The resolution of UFAM is significantly higher than that of agarose microspheres, and the peak shape of the elution peak is more "sharp".


Good pH, reductant and chelators tolerance

UFAM is stable from pH 1 to 14 (pH 3~13 is recommended for prolonged use), so it can be cleaned in position (CIP) with 0.5 M NaOH or even higher base concentrations. More efficient removal of heat sources as well as hetero-proteins.

  • Resistant to 30 mM EDTA (> 48 hours)
  • Resistant 20 mM DTT and (> 48 hours)
  • 45 mM β-mercaptoethanol (> 48 hours)
  • Compatible with any samples that mental chelators can work with.
