Application:
Preparation of Fine Culture Medium
Materials: Dehydrated media powder, distilled or MQ water.
Procedure:
- Dissolve the specified amount of dehydrated media powder and desired amino acids powder in distilled or deionized water.
- Follow the manufacturer's recommendations for the amount of powder and water.
- Mix thoroughly and heat if necessary to ensure complete dissolution.
- Autoclave at 121°C for 15 minutes.
- Allow the medium to cool before use.
Nutrition Deficit Strain Selection Media
Materials: Basal media, specific nutrients (e.g., amino acids, vitamins).
Procedure:
- Prepare the basal media by dissolving the required ingredients in distilled water.
- Adjust the pH if necessary.
- Add a specific set of amino acids to create a selective environment for the desired strain.
- Autoclave.
- Pour the media into sterile Petri dishes or tubes and allow it to solidify.
Preparation of AAs Stock Solution for CFPS Reaction System
Materials: Amino acids, MQ water(RNase-free water or DEPC treated water), buffer solution( HEPES, TES or MOBS).
Procedure:
- Weigh the required amount of each amino acid.
- Dissolve the amino acids in distilled water or a suitable buffer solution.

- Filter-sterilise the solution using a 0.22 µm filter.
- Store the stock solution at -20°C until use.
