Yeast Peptone Dextrose Adenine (YPDA) medium, 250g
Universal media for yeast proliferation culturing (enrichment). YPDA is the same as YPD but with the addition of an adenine [1] which prevents yeast strains with ADE1 and ADE2 mutations [2] from turning pink/red after prolonged growth.
Per 1000mL of YPDA medium contains
Yeast extract 10g
Peptone 20g
Glucose 20g
Adenine sulphate 30 mg
Preparation
- Weigh 50 g YPDA [3] and dissolve in 800 ml ddH2O or MQ water by stirring. Add 15~20 g agar at this stage if YPDA/agar plates are being prepared. Make it 1000 ml with water.
- Autoclaving at 116℃ for 30 minutes is recommended for sterilisation.
[Option] Aseptically add an appropriate volume of filter-sterilised antibiotic stock once the media has cooled down.
- Unused prepared media can be stored at 4°C, the shelf life is about several months.
[3] For preparing 2X YPDA Broth, dissolve 100.06 g of YPDA medium powder. For preparing 1/2 YPDA broth, dissolve 25 g of YPDA medium powder.
[1] The regular YPDA formula contains adenine at a final concentration ranging from 30 to 50mg/L. Higher concentrations may help extend yeast cultivation, whilst concentrations above 120mg/L sometimes have repressive effects on yeast growth.
